Острые респираторные инфекции (ОРИ) являются одной из основных причин заболеваемости и смертности от инфекционных заболеваний в мире. Респираторные инфекции могут быть вызваны патогенами различной природы: вирусами, бактериями, микоплазмами и др. Быстрая и точная идентификация в биологических образцах патогенов, таких как бактерии является важной задачей, для которой используется секвенирование гена 16S рРНК с применением платформ нового поколения.</p><p>Цель – сравнительный анализ качественных характеристик микробиома ротоглотки здоровых добровольцев и больных ОРИ неустановленной этиологии по данным секвенирования гена 16S рРНК.</p></sec><sec><title>Методы и материалы</title><p>Методы и материалы. Получены и проанализированы данные секвенирования MiSeq Illumina региона V3–V4 16S рРНК мазков из ротоглотки госпитализированных пациентов с симптомами ОРИ и здоровых пациентов.</p></sec><sec><title>Результаты</title><p>Результаты. Был проведен анализ данных секвенирования области V3–V4 16S рРНК у 116 госпитализированных пациентов с симптомами ОРИ и 81 здорового пациента. У пациентов с ОРИ наблюдалось более высокое содержание условно-патогенной микрофлоры, в частности Staphylococcus, Ralstonia, Aeribacillus, Acinetobacter baumannii, Methylobacterium-Methylorubrum, Rhodococcus equi. В контрольных образцах преобладала нормальная комменсальная микробиота дыхательных путей, такая как Neisseria, Prevotella, Fusobacterium, Veilonella.</p></sec><sec><title>Выводы</title><p>Выводы. В образцах микробиоты госпитализированных пациентов с ОРИ обнаружено преобладание оппортунистической и потенциально патогенной микробиоты, в то время как у здоровых добровольцев преобладают нормальные представители микробиоты дыхательных путей. Для более детального анализа необходимы данные о видовом составе микробиоты, которые можно получить при секвенировании полной последовательности гена 16S рРНК.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Introduction</title><p>Introduction. Acute respiratory infections (ARI) are one of the main causes of morbidity and mortality from infectious diseases in the world. Respiratory infections can be caused by pathogens of various etiologies: viruses, bacteria, mycoplasmas, etc. Rapid and accurate identification of pathogens, such as bacteria, in biological samples is an important task, for which 16S rRNA gene sequencing using new generation platforms is used.</p><p>The objective was a comparative analysis of the qualitative characteristics of the oropharyngeal microbiome of healthy volunteers and patients with ARI of unknown etiology based on the 16S rRNA gene sequencing.</p></sec><sec><title>Methods and materials</title><p>Methods and materials. Using V3–V4 region of 16S rRNA Illumina MiSeq sequences from oropharyngeal swabs, we analyzed the microbiome of hospitalized patients with ARI symptoms and healthy patients.</p></sec><sec><title>Results</title><p>Results. In this study, we conducted V3–V4 region of 16S rRNA sequencing analyses of the oropharyngeal samples from 116 hospitalized patients with ARI symptoms and 81 healthy patients. Patients with ARI exhibited higher abundance of opportunistic pathogens, particularly Staphylococcus, Ralstonia, Aeribacillus, Acinetobacter baumannii, Methylobacterium-Methylorubrum, Rhodococcus equi. In the control samples, normal commensal respiratory tract microbiota, such as Neisseria, Prevotella, Fusobacterium, Veilonella was dominated.</p></sec><sec><title>Conclusions</title><p>Conclusions. The microbiota samples of hospitalized patients with ARI showed a predominance of opportunistic and potentially pathogenic microbiota, while normal representatives of the respiratory tract microbiota predominate in healthy volunteers.
